Background
Mammalian cells express two structurally redundant kinases (Cdk4 and Cdk6) and three structurally redundant partner cyclins (D1, D2 and D3), with predominant expressions of Cdk4 and D1 in mesenchymal cells and Cdk6 and D3 in hematopietic cells. Cdk6 is though to be functionally redundant with Cdk4 and to play the same role in hematopoietic cells as does Cdk4 in fibroblasts. Cyclin D3 is widely expressed in a variety of cells and has been suggested to play a role in the G1-S transition of hematopoietic cells and fibroblasts. Furthermore, recent studies have implicated the involvement of this cyclin subtype in a variety of decisions on cell fate, including proliferation, apoptosis and differentiation.
